How to approach them
Look at the row and column sharing the tightest empty square. Compare the shapes in the current tray before placing one there, and keep the remaining gaps usable for later pieces.

Do the block colors have to match to clear a line?
No. A filled row or column clears regardless of the colors used. Shape and placement are what matter.
